Aptamers as diagostic tools
The inventions provide aptamers as diagnostic tools for detection of APC in biological samples also in the presence of abundant inactive protein C with high selective specificity. This allows establishment of routine diagnostics for APC to analyse disturbances in haemostasis, e.g. in sepsis or in haemophilia A. Further the inventions provide therapeutic aptamers binding to APC for the control of bleeding events, e.g. in haemophilia A or Willebrand syndrome.
Inhibition of thrombin-induced platelet aggregation
In another embodiment of the inventions, linkage of aptamers HD1 and HD22 by a poly-dA-linker generates a fusion aptamer, named HD1-22, that binds with higher affinity to thrombin than the original aptamers.
The determined effect of HD1-22 on thrombin-induced platelet aggregation using washed human platelets showed superior inhibitory activity as an equimolar mixture of both precursor aptamers. Under the conditions tested, HD1-22 was found two-fold more effective as bivalirudin and »5-fold more active as argatroban.
Photo-activated aptamers
Additionally the inventions comprise aptamers binding to haemostatic target molecules, such aptamers being activated to covalently bind to the target molecules by application of light. Such photo-activated aptamers could be used for example for the control of bleeding and occlusion in surgical procedures.
