Building on the pioneering work at EMBL, Heidelberg, on Estrogen signaling and allosteric kinase inhibition, ELARA has four chemotypes in lead optimization with a broad indication range and the potential of lower toxicity due to their unique modes of action.

Their most advanced candidate is orally bioavailable and indicated for the treatment of multiple myeloma, a cancer that clearly lacks efficacious therapies. ELARA foresees an IND filing by Q1 2010 and initiation of human proof of concept trials.

ELARA has developed a robust pre-clinical pipeline with sets of promising molecules. These are directed at a number of clinically validated and also novel targets like Aurora kinases and cellular factors involved in Estrogen Receptor signaling, which are expected to open new therapeutic perspectives in oncology.

ELR105813

is an orally bioavailable tubulin polymerization inhibitor targeting the colchicine binding site. It has a broad spectrum of activity in vitro against a diverse set of tumour cell lines. ERL105813 is well tolerated in vivo and shows superior pharmacokinetic profile compared to ABT-571, a phase II compound with similar mechanism of action. Xenograft studies are ongoing and initial data is expected by end of 2007.

ELR510052

a second chemotype targeting the colchicine binding site of tubulin. They have reason to believe that this second generation of tubulin inhibitors will show enhanced in vivo profile compared to the competition. Proof of efficacy results are expected in Q1 2008.

ERL144949

targets tubulin polymerization through an indirect mechanism. This chemotype show good levels of in vitro activity against a diverse set of tumor cell lines. Current efforts are focused on optimization for physicochemical properties.

Allosteric Kinase Inhibitors: Aurora

Ground breaking research from EMBL research groups enabled the identification of a novel allosteric regulatory site of Aurora A kinase that is essential for proper protein function. In our Aurora program, we established a proprietary portfolio of allosteric inhibitor chemotypes.
