Species-independent detection method - nested SAPD-PCRSummary
species-independent detection method that allows the determination of the genetic relationship of arbitrary organismsOrganization name
IMG Innovations-Management GmbHProfile
The analysis of the DNA and the genome respectively is the state-of-the-art method for the detection of the identity or the relationship between different individuals.
In addition to highly-conserved regions that are stable within one art, there are also variable regions (polymorphisms) in the genome of organisms. The differences in the sequence of the DNA may be due to the exchange, the insertion or the deletion of single bases or whole segments of DNA. Fingerprintmethods allow the genome typing of species, strains or serovars if there are only few genomic sequencedata available.
The known methods, as for example the RAPD-PCR, have in common that often they do not provide reliable or reproductive information about the degree of relationship or the derivation of individuals with small genetic distance or that they cover only a small segment of organisms. Therefore the supply of a new detection method will entail essential advantages in modern biology and medicine.
Application
The SAPD-PCR (Specific amplified polymorphic DNA-PCR) is a species-independent detection method that allows the determination of the genetic relationship of arbitrary organisms. The method can be applied as well to human beings as to animals, plants or microorganisms and can therefore be used for the determination of species, strains or serovars.
Advantages
- Rapid and simple viability as compared to the existent procedures (RFLP-PFGE)
- High reproducibility
- Low costs of material
- Application for arbitrary groups of organisms
