Several isoquinoline alkaloids with interesting, pharmaceutically useful properties have recently been isolated from marine as well as terrestrial organisms. They served as models for 3,3-dimethyl-8-oxo-di-(or: tetra-)hydroisoquinolines, a compound class which is easily accessible by total synthesis and for which pronounced activities against protozoae have been found in cell vitality assays.

Advantages

  • Simple synthetic procedure giving high yield; compatible with various substitution patterns and therefore appropriate for the synthesis of larger compound libraries
  • High in vitro activity of the lead structure (IC50 = 32 nM)
  • Significant specificity against selected protozoae; almost immeasurable cytotoxicity toward mammalian cells
  • Partially additional activity against leishmaniasis or antiviral activity (for plasmodial secondary infection as a result of low immunity, e.g. after HIV primary infection)  
  • Expectedly good "drug likeness" due to structural analogy with low-molecular-weight natural compounds
