Recently several natural compounds with anti-tumour effects have been isolated from tunicates, sponges, as well as marine microorganisms of which some have already found their way into advanced clinical trials.
The physiological role of natural products with isoquinoline or naphthalene skeletal frameworks served as a model for the three synthesised compound classes A-C, for which for the first time a pronounced growth inhibition of various tumour cell lines was now found:
- Oxotetrahydro-isoquinolines (A.)
- Phenyl-aminomethyl-naphthalenes (B.)
- Alkylthio-naphthoquinones (C.)
Advantages
- Simple synthetic chemical procedures giving high yields; compatible with various substitution patterns and therefore appropriate for the synthesis of larger compound libraries
- As low-molecular-weight natural compounds (C) or their structural analogues (A,B) expectedly good "drug likeness"
- Distinct in vitro activities of the lead structures (e.g. ED50 = 340 nM for C) with uniformly broad activity profiles
- Almost immeasurable cytotoxicities of the lead structures and many derivatives toward healthy mammalian cells (A, B)
- Minimal sub-acute toxicity in animal experiments (C)
- Twofold mode of action: simultaneous inhibition of cell proliferation and angiogenesis (C)
- Additional antiviral activity - if an infection by tumor viruses preceded (A)
Stage of Development
- Scaleable procedures to chemically synthesize the pharmaceutical agents - also in enantiomerically pure form
- Procedure to isolate the pharmaceutical agents from microorganisms (C)
- Quantitative structure-activity profiles of a multitude of variously substituted derivatives against different tumor cell lines, in particular mouse lymphoma cells
- Rational model of function within the marine organism based on molecular biological studies (C)
