Patients with distant metastasized melanomas have a negative prognosis and a low survival rate. Current therapies are surgery in dependence of metastasis migration as well as the application of combined chemotherapies. Latest clinical studies deal with cytokine therapies such as interleukin-12 and interferon-gamma but without significant success in comparison to traditional methods. It is therefore possible that beside a cytokine milieu a set of co-stimulatory-surface antigen expression or repression on the professional antigen presenting cells are required to break the tolerance to melanomas.
IFN-y; and IL-12 are also considered as a protective cytokine milieu counteracting the CD4+Thelper type 2 cytokines involved in the onset and development of allergic asthma. The traditional medication results in the application of corticosteroids, bronchodilators and antihistamines. These drugs are used to medicate the symptoms of asthma but they have no effect on the onset of this wide spread disease. Unfortunately single cytokine delivery such as IL-12 has not been shown to be successful either, so that new therapeutical approaches must be considered in the treatment of allergic asthma
