DNA typing of distinct STR loci establishes the identification of individuals or biological samples collected at crime scenes since several years. If only low amounts of DNA are available and /or the quality of the DNA is rather poor due to degradation, with commercial multiplex kits the typing often results in no or only partial DNA profiles.
The innovation comprises a method in which two PCR reactions (a sixplex and a fiveplex PCR) are combined into one large multiplex reaction. After amplification the preparation is splitted and specifically labelled amplicons are further analysed.
Application
The innovative BioPlex-11 multiplex PCR system provides a significant and sensitive system for a powerful analysis of degraded and low copy DNA samples. It allows the simultaneous analysis of ten short amplicon STR systems from a single aliquot.
Advantages
- Samples were successfully typed down to 25 pg of DNA (equivalent of approx. four cell nuclei) demonstrating the high sensitivity of the assay
- Correct genotypes were obtained for highly degraded DNA samples
- The assay could be applied for analyses of telogen hair roots and for genotyping of tsunami cases

- Specificity and reproducibility of the method was verified by testing 15 blood samples
- Application of the assay in practical work of analysing telogen hair roots from real casework
